Wherever two straight links intersect, it will be an intersection.

I am wondering if there is a way to define at-grade separation where two straight link intersects?

My best guess is "NO". But really want to confirm with you.

your best guess is wrong, if you use Version 6 Smile.
if you add one link with curvature it will not be an intersection with crossing straight/curved links. even if you remove the curvature from that link it would still not be an intersectin. add a node in one of the link and put some number in Z elevation field in the properties of the node. then see the simtraffic simuation, you have your grade separation.

To add to this topic, hold your [Ctrl] key when crossing links to create grade separation. This works for straight links.
